1: What are the safety precautions for high risk TB labs?
Solution ;:
The formation of aerosols in high risk TB labs is very, because in these labs high concentrations are
available. That put the health of lab workers in risk. That’s why world health organization issued some
safety precautions which are required to follow;
These are…
❖ The lab should be double door. One fix entry and one fix exit door. At one time, one door should
be allowed to open.
❖ All the hazard materials should be labeled as biohazard symbol.
❖ The workers should wear Lab coat, gloves, face mask lab cape, protection glasses and show
cover.
❖ Before starting experiments all the equipments used in High risk TB test should be
decontaminate with 70% alcohol.
❖ Before leaving the lab dispose the gloves, face mask, lab cape and show cover. Clean the
protective glasses in safe place.
2: What is the mode of action of pUC18 restriction enzyme?
Solution ;
The pUC18 is made of br322 and one strand of M13. The br322 have amp r site which is resistance to ampicillin. The M13 region consist of promoter and modified sequence polylinker. The pUC18 enzyme hydrolyzed the polylinker and that way this polylinker received DNA
sequence.
